Connecting the terms "colorimeter tube", "metal bath" and "spectrophotometer", a complete and classic quantitative analysis process in the laboratory is described. They respectively play the roles of reaction container, reaction condition control device and final detection instrument.
In simple terms, this is a complete chain of "sample pretreatment → color reaction → quantitative detection".

The relationship and workflow of the three
Colorimeter tube (reaction container)
Role: The "stage" for chemical reactions.
Function: Used for precisely measuring and mixing the sample to be tested and the color-developing reagent. Many standard detection methods (such as water quality testing, food safety testing) specify the use of capped colorimeter tubes for reactions because it can prevent the liquid from evaporating when heated and facilitate the observation of color changes.
Metal bath (reaction condition control)
Role: The "precise environmental controller".
Function: Provides a constant and uniform temperature for the color reaction in the colorimeter tube. Many chemical reactions (especially enzyme reactions or reactions that require high-temperature digestion) are extremely sensitive to temperature. The metal bath can ensure that all samples undergo reactions at exactly the same temperature, which is crucial for ensuring the accuracy and repeatability of experimental results.
Spectrophotometer (quantitative detection)
Role: The "final data reader".
Function: After the reaction is completed, transfer the solution in the colorimeter tube to a more precise cuvette and place it in the spectrophotometer. The instrument emits light of a specific wavelength through the solution and, according to the Lambert-Beer law (A = εcL), measures the absorbance (A) of the solution to precisely calculate the concentration (c) of the analyte being tested.

 Example of standard operating procedure
The steps for a typical experiment using these three are as follows:
Preparation for reaction: In several clean colorimeter tubes, add standard solutions and the sample to be tested, and then add the same amount of color-developing reagent.
Constant temperature color reaction: Place all the colorimeter tubes in the metal bath set at a specific temperature, heat or keep them at a certain temperature for a period of time to allow the color reaction to proceed fully and stably.
Cooling and transfer: After the reaction is complete, remove the colorimeter tubes and allow them to cool to room temperature. Then transfer the color-developed solution to a cuvette.
Instrument measurement: Use the spectrophotometer, first zero the instrument with a blank control solution (blank correction), and then measure the absorbance values of each sample successively.
Data analysis: Draw a standard curve based on the absorbance and concentration of the standard solution, and then find the concentration of the sample on the curve.
